🚨 New paper in Nature Communications: Structure-guided design yields a sub-nanomolar, human-selective P2X7 antagonist
I am delighted to share our latest publication, “A polycyclic scaffold identified by structure-based drug design effectively inhibits the human P2X7 receptor,” now published in Nature Communications. The study combines high-resolution cryo-EM, functional pharmacology, and molecular dynamics to uncover human-specific structural features of the P2X7 receptor and to design UB-MBX-46, a highly selective, sub-nanomolar antagonist of human P2X7.

🧠 In this study, we uncover a novel pharmacological action of memantine, a well-known FDA-approved drug for Alzheimer’s disease. Beyond its established role as an NMDA receptor antagonist, we show that memantine also inhibits calcium-permeable AMPA receptors (CP-AMPARs)—key players in neurodegenerative and neurodevelopmental disorders.

The Medicinal Chemistry team stepped away from the lab for a well-deserved day of fun and relaxation in Castelldefels. We enjoyed a variety of ball games and concluded the day with a cheerful shared lunch.
